    Overview 本試劑盒利用Taqman探針定量PCR方法,能快速檢測各種生物制品及藥品的中間品、半成品和成品 中殘留的中國倉鼠卵巢(CHO)宿主細胞DNA。試劑盒中引入了尿嘧啶-N-糖基化酶(uracil-N-glycosylas e, UNG)防污染系統(tǒng),可有效去除PCR產(chǎn)物的殘留污染,大大降低了由于擴增產(chǎn)物污染而導致的假陽性。 檢測體系設(shè)置為雙通道系統(tǒng),組分中含有內(nèi)標控制(IC),通過IC的信號表現(xiàn),監(jiān)控反應(yīng)過程是否正常 進行,排除假陰性現(xiàn)象。 本產(chǎn)品適用于以中國倉鼠卵巢細胞(如CHO-K1, CHO-S, CHO-DG44, CHO-GS等細胞系)為宿主生產(chǎn) 的生物藥原液、成品、半成品、中間品或工藝過程監(jiān)控樣本。為獲得定量樣本DNA的校準曲線,本試劑 盒需要制備6個稀釋梯度的校準品(300 pg/μL到3 fg/μL),試劑盒中包含制備校準曲線的DNA校準品CHO DNA Control,溯源至CHO細胞DNA含量測定國家標準品(中國,編號:270026)。 CHO宿主細胞的殘留DNA抽提建議使用本公司配套的通用型宿主DNA殘留樣本前處理試劑盒。首次 使用前建議先完成產(chǎn)品的適用性研究:確認樣本基質(zhì)是否存在干擾;確定合適的樣本檢測稀釋條件;樣 本中是否存在反應(yīng)抑制物等。針對質(zhì)?;蛘哳愃坪怂針颖?,尤其需要考察產(chǎn)品適用性,確認樣本干擾以 及優(yōu)化檢測方案。

    ApplicationIn the production and purification process of antibody products, Protein A affinity chromatography resin is a commonly used tool to separate and purify antibodies. However, during the antibody purification process, Protein A will fall off, causing antibody products contains Protein A, thus affecting the purity and potency of the product, causing major problems to the pharmaceutical industry. For example, the 2020 edition of the Chinese Pharmaceutical stipulates that the Protein A residue detected by enzyme-linked immunoassay for nimotuzumab injection should not be higher than 0.001% of the total protein, so the Protein A residue in biological products must be detected.
