產(chǎn)品概述
粒細(xì)胞巨噬細(xì)胞集落刺激因子(GM-CSF)是一個(gè)多效性的細(xì)胞因子,對(duì)造血細(xì)胞有多方面的影響。它可以將CD34+前體細(xì)胞動(dòng)員到外圍從而促進(jìn)它們增殖、存活以及分化成中性粒細(xì)胞、單核細(xì)胞/巨噬細(xì)胞、嗜酸性細(xì)胞和骨髓樹突狀細(xì)胞。GM-CSF對(duì)這些終末分化的骨髓細(xì)胞效應(yīng)功能的誘導(dǎo)也是必需的。此外,GM-CSF對(duì)于紅細(xì)胞和巨核細(xì)胞的增殖和分化也起到促進(jìn)作用。GM-CSF由一系列不同的細(xì)胞產(chǎn)生,包括角化細(xì)胞、成熟和未成熟的NK細(xì)胞、II型肺泡上皮細(xì)胞、內(nèi)皮細(xì)胞、單核細(xì)胞、骨髓間葉干細(xì)胞,CD4+和CD8+T細(xì)胞、巨核細(xì)胞、B細(xì)胞、嗜酸性細(xì)胞、軟骨細(xì)胞和纖維原細(xì)胞。
產(chǎn)品原理
本實(shí)驗(yàn)采用雙抗體夾心ELISA法??谷薌M-CSF單抗包被于酶標(biāo)板上,標(biāo)本和標(biāo)準(zhǔn)品中的GM-CSF會(huì)與單抗結(jié)合形成免疫復(fù)合物,游離的成分被洗去;加入生物素化的抗人GM-CSF抗體,生物素化抗人GM-CSF抗體與酶標(biāo)板上結(jié)合的標(biāo)準(zhǔn)品或樣本中的GM-CSF結(jié)合而形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標(biāo)記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有GM-CSF,辣根過氧化物酶會(huì)使無色的顯色劑變藍(lán),加終止液變黃。在450nm處測OD值,GM-CSF濃度與OD450值之間呈正相關(guān),可通過繪制標(biāo)準(zhǔn)曲線求出標(biāo)本中GM-CSF濃度。
