產(chǎn)品概述

細胞間粘附分子-1（Intercellular adhesion molecule-1，ICAM-1），也叫做CD54，是一個普遍存在的跨膜糖蛋白，在白細胞的遷移和激活中起著非常重要的作用?？扇艿腎CAM-1被報道出現(xiàn)在血清、腦脊髓液、尿液和支氣管肺泡的洗出液中。在這些液體中 sICAM-1（Soluble ICAM-1）量的提高與心血管疾病、II型糖尿病、器官移植失功、氧化劑應(yīng)激、腹部脂肪聚集、高血壓、肝病和一些惡性腫瘤密切相關(guān)。sICAM-1促進血管生成并且可以作為血管內(nèi)皮細胞激活和受損的一個指標。sICAM-1也可以作為跨膜ICAM-1介導(dǎo)活性的抑制劑，比如抑制單核細胞粘附到激活的內(nèi)皮細胞，抑制腫瘤細胞對NK細胞介導(dǎo)的裂解的敏感性。

產(chǎn)品原理

本實驗采用雙抗體夾心ELISA法?？谷薎CAM-1單抗包被于酶標板上，同時加入標本、標準品和生物素化的抗人ICAM-1抗體，標本、標準品中的ICAM-1會與加入于孔中的生物素化的抗人ICAM-1抗體及包被于酶標板上的單抗結(jié)合，形成免疫復(fù)合物，游離的成分被洗去；加入辣根過氧化物酶標記的親合素，親合素與生物素特異性結(jié)合，游離的成分被洗去。加入顯色底物（顯色劑），若反應(yīng)孔中有ICAM-1，辣根過氧化物酶會使無色的顯色劑變藍，加終止液變黃。在450nm處測OD值， ICAM-1濃度與OD₄₅₀值之間呈正相關(guān)，可通過繪制標準曲線求出標本中ICAM-1濃度。

產(chǎn)品性能

1. 靈敏度：最小可測人sICAM-1達100pg/ml 。

2. 重復(fù)性：板內(nèi)、板間變異系數(shù)均<10%。

產(chǎn)品組分及儲存條件

試驗所需自備儀器和試劑:

1. 酶標儀(450nm檢測波長濾光片，570nm或630nm校正波長濾光片)。

2. 高精度加液器及一次性吸頭：0.5-10μl，2-20μl，20-200μl，200-1000μl。

3. 37℃恒溫箱、蒸餾水或去離子水、微量振蕩器、坐標紙。
