產(chǎn)品概述
細胞間粘附分子-1(Intercellular adhesion molecule-1,ICAM-1),也叫做CD54,是一個普遍存在的跨膜糖蛋白,在白細胞的遷移和激活中起著非常重要的作用??扇艿腎CAM-1被報道出現(xiàn)在血清、腦脊髓液、尿液和支氣管肺泡的洗出液中。在這些液體中 sICAM-1(Soluble ICAM-1)量的提高與心血管疾病、II型糖尿病、器官移植失功、氧化劑應(yīng)激、腹部脂肪聚集、高血壓、肝病和一些惡性腫瘤密切相關(guān)。sICAM-1促進血管生成并且可以作為血管內(nèi)皮細胞激活和受損的一個指標。sICAM-1也可以作為跨膜ICAM-1介導(dǎo)活性的抑制劑,比如抑制單核細胞粘附到激活的內(nèi)皮細胞,抑制腫瘤細胞對NK細胞介導(dǎo)的裂解的敏感性。
產(chǎn)品原理
本實驗采用雙抗體夾心ELISA法??谷薎CAM-1單抗包被于酶標板上,同時加入標本、標準品和生物素化的抗人ICAM-1抗體,標本、標準品中的ICAM-1會與加入于孔中的生物素化的抗人ICAM-1抗體及包被于酶標板上的單抗結(jié)合,形成免疫復(fù)合物,游離的成分被洗去;加入辣根過氧化物酶標記的親合素,親合素與生物素特異性結(jié)合,游離的成分被洗去。加入顯色底物(顯色劑),若反應(yīng)孔中有ICAM-1,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值, ICAM-1濃度與OD450值之間呈正相關(guān),可通過繪制標準曲線求出標本中ICAM-1濃度。
